SUMMARY:Predicting Human Pharmacokinetics and Doses of Diverse Modalities: The Current State of the Art and Case Studies
DESCRIPTION:This PBSS workshop will present cutting-edge tools\, methods\, and case studies for predicting human pharmacokinetics (PK) and dose selection across various modalities\, including small molecules\, oligonucleotides\, and biologics. Topics include machine learning-based PK prediction\, cross-industry efforts from the IQ Consortium\, and applications to emerging modalities such as siRNAs and PROTACs. The workshop will also cover the role of pharmacokinetic/pharmacodynamic (PK/PD) relationships in guiding first-in-human dose selection\, with case studies on monoclonal and bispecific antibodies\, as well as immunomodulators informed by HESI’s work beyond the minimal anticipated biological effect level (MABEL). A final case study will demonstrate the integration of PK/PD modeling with nonclinical data to support therapeutic index estimation. Attendees will gain practical insights into advancing predictive strategies for drug development of diverse modalities. \nREGISTER» \n

SUMMARY:Technology: A Drug Repurposing Approach for Clinical Treatment of Triple-Negative Breast Cancer
DESCRIPTION:Register for a free NIH technology webinar to learn about a potential\, novel treatment for Triple Negative Breast Cancer (TNBC). William Reinhold and his team at the National Cancer Institute repurposed two separate therapeutic compounds ─ acetalax and bisacodyl (both previously used as laxatives) ─ into novel treatments for TNBC. Attend to learn more about this technology and opportunities to help develop and commercialize it. \nAbout the Technology\nThe Problem:\nTriple negative breast cancer (TNBC) is a highly invasive and hard to treat cancer. Responses to current treatment methods are usually short lived\, followed by rapid relapse leading to visceral and brain metastasis. There is also no standard chemotherapy regimen for patients with relapsed/refractory TNBC.  Therefore\, there is a significant unmet need for new and effective treatment options to treat patients with TNBC. \nHow the Solution Works:\n\nAcetalax and bisacodyl show promising results in killing TNBC cells in patient-derived xenografts (PDX) cell lines and mouse models.\nAcetalax and bisacodyl disrupt the activity of the protein transient receptor potential melastatin member 4 (TRPM4)\, which is over expressed in TNBC and several other cancers.  \nDisruption of TRMP4 activity negatively affects its ability to regulate cancer cell migration\, cytoskeleton maintenance and epithelial-mesenchymal transition (EMT). \nAcetalax and bisacodyl’s affect on TRMP4 disrupts the cancer cell membrane\, causing oncosis (i.e.\, swelling and rupture)\, which kills cancer cells.\n\nCompetitive Advantages\n\nRepurposed drugs provide reduced development costs and reduced development timelines\nBisacodyl may be eligible for expedited FDA approval based on current approval for different indication\nPotential novel TNBC treatment option compared to the current challenging to treat standard of care\n\nWho should attend?\n\nBusiness development professionals\nScientific and drug development professionals\nBiotech/pharma/academia researchers\nInvestors and entrepreneurs\n\nWhy attend?\n\nAssess co-developing the technology\nInteract with the inventor\, ask questions and provide feedback\nLearn how to partner with the NIH facilitated by the NCI Technology Transfer Center\n\nIf you are interested in learning more about how to co-develop and/or license this technology\, please contact Michael.Salgaller@nih.gov. \n\n\n\n\n\n\n\n\n\n\n\n\n\n\nREGISTER»
